Provided is a method for measuring the diffusion characteristic value (e.g., the diffusion coefficient) of a light-emitting particle by using the scanning molecule counting method that employs optical measurement with a confocal microscope or multiphoton microscope. The method of the present invention for measuring the diffusion characteristic value of a light-emitting particle is characterized by measuring the strength of a light beam to create optical intensity data while the optical path of optics in a microscope is changed to thereby allow the position of an optical detection region of the optics to be periodically moved in a sample solution along a predetermined path; and calculating the diffusion characteristic value of the light-emitting particle on the basis of a shift in time of the interval between points in time, at which a plurality of signals occur corresponding to the same light-emitting particle on the optical intensity data, from the travel cycle of the optical detection region.
