||PCT/RU2014/000941||MAMONTOVA, Marina Vasilevna МАМОНТОВА, Марина Васильевна||POSPELOV, Vadim Igorevich ПОСПЕЛОВ, Вадим Игоревич|
The invention relates to biotechnology, and specifically to a method for isolating microRNA from biological fluids which contain exosomes. The method involves subsequent centrifugation, ultrafiltration and ultracentrifugation of a condensed culture medium, dissolving the produced precipitate in a phosphate salt buffer and further centrifugation. The produced precipitate is dissolved in water, and the solution is subjected to free flow electrophoresis using an FFE system apparatus. Each produced fraction then undergoes ultracentrifugation. Each fraction is cleaned of cell walls of exosomes by means of filtration and centrifugation, and a reverse transcription reaction is carried out using reverse transcriptase. The reaction is stopped by heating to 95°С over a period of 5 minutes.
