(EN)Affinity chromatography media are prepared by reacting mono or di-chloro triazine or related dyes with a solid support matrix containing free hydroxy or amine groups in the presence of an alkali metal hydroxide. In the absence of cyanogen bromide activation, the linkage to the support is entirely via the triazine ring giving high and specific protein-binding capacity. Suitable support matrices include polymers and copolymers of dextrose, dextran and amides and especially agarose. Celluloses may also be used. Dye binding levels are much higher than those obtained in the presence of carbonates. Mono-chlorotriazinyl dyes are bound at a pH preferably above 9.5 for about 40-60 hours at room temperature or less at elevated temperatures. Dichlorotriazinyl dyes are bound at pH 8 to 12.5 in a few hours at room temperature. The media may be used for the efficient and often highly specific purification of a wide variety of proteins, including enzymes and blood proteins.
(FR)On prepare des milieux de chromatographie par affinite en faisant reagir une mono ou une dichloro triazine ou des colorants apparentes avec une matrice support solide contenant des groupes hydroxy ou amine, en presence d"un hydroxyde de metal alcalin. En l"absence d"activation par le bromure de cyanogene la liaison au support s"effectue entierement par le noyau de la triazine et confere une capacite de liaison des proteines elevee et tres specifique. Parmi les matrices support qui conviennent on peut citer les polymeres et les copolymeres de dextrose, de dextrane et d"amides, notamment l"agarose. On peut egalement utiliser des celluloses. Les capacites de liaison des colorants sont beaucoup plus elevees que celles qui sont obtenues en presence de carbonates. Les colorants monochlorotriazinyle sont lies a un pH de preference superieur a 9, 5 pendant environ 40 a 60 heures a temperature ambiante ou pendant une duree plus courte a des temperatures plus elevees. Les colorants dichlorotriazinyle sont lies a un pH de 8 a 12, 5 pendant quelques heures a temperature ambiante. Les milieux peuvent etre utilises pour la purification efficace et souvent tres specifique d"une large variete de proteines, y compris les enzymes et les proteines du sang.
