Abstract
(EN) A kit for the detection of virus particles using immune molecules. A monoclonal antibody of a virus envelope antigen is subjected to biotin modification, and a magnetic bead is coupled with streptavidin; then, the biotin-modified monoclonal antibody is incubated with a virus-containing solution, the antibody and virus particles or an antigen form a complex, then, the streptavidin coupled magnetic bead is added for incubation, and the streptavidin on the magnetic bead binds, with high specificity and high affinity, to the biotin coupled antibody so as to specifically capture a virus particle with the envelope; and after a supernatant is separated by using a magnetic separator, the complete virus particle, an empty capsid virus and a free envelope antigen can be separated from other virus components, and then the magnetic bead conjugate is qualitatively or quantitatively detected by means of PCR amplification.
(FR) L'invention concerne un kit pour la détection de particules virales à l'aide de molécules immunitaires. Un anticorps monoclonal d'un antigène d'enveloppe virale est soumis à une modification à la biotine, et une bille magnétique est couplée à la streptavidine; puis, l'anticorps monoclonal modifié à la biotine est incubé avec une solution contenant un virus, l'anticorps et les particules virales ou un antigène forment un complexe, puis, la bille magnétique couplée à la streptavidine est ajoutée pour l'incubation, et la streptavidine sur la bille magnétique se lie, avec une haute spécificité et une haute affinité, à l'anticorps couplé à la biotine de façon à capturer spécifiquement une particule virale avec l'enveloppe; et après la séparation d'un surnageant à l'aide d'un séparateur magnétique, la particule virale complète, un virus à capside vide et un antigène d'enveloppe libre peuvent être séparés des autres composants du virus, puis le conjugué à bille magnétique est détecté qualitativement ou quantitativement au moyen d'une amplification par PCR.
(ZH) 一种免疫分子病毒颗粒检测试剂盒,将病毒包膜抗原的单克隆抗体进行生物素修饰,将磁珠与链霉亲和素进行偶联;然后将生物素修饰的单克隆抗体与含有病毒的溶液进行孵育,抗体与病毒颗粒或抗原形成复合物,然后加入已经偶联链霉亲和素的磁珠进行孵育,磁珠上的链霉亲和素将与偶联了生物素的抗体进行高特异高亲和力结合,进而特异性地捕获具有包膜的病毒颗粒,再通过磁分离器分离上清液后,完整病毒颗粒及空壳病毒、游离的包膜抗原可以与其它病毒组分分离,然后通过PCR扩增对磁珠结合物进行定性或者定量检测。
