Abstract
(EN) The disclosure relates to a recombinant DNA dependent DNA polymerase having 5'-3' exonuclease activity and lacking 3'-5' exonuclease activity, wherein said polymerase is capable of extending DNA polymerisation from a single mismatched base pair and has an error rate of at least 1:1000. The disclosure further relates to a nucleic acid molecule encoding the recombinant DNA dependent DNA polymerase, a method for synthesizing double stranded DNA (dsDNA), a method for obtaining the position of a single strand break in a template dsDNA molecule, a method to introduce mutations in DNA of bacterial or eukaryotic cells, or organisms.
(FR) La présente divulgation concerne une ADN polymérase recombinée dépendante de l'ADN ayant une activité de exonucléase de 5' à 3' et dépourvue d'activité d'exonucléase de 3' à 5', ladite polymérase étant capable d'étendre la polymérisation de l'ADN à partir d'une seule paire de bases mésappariées et ayant un taux d'erreur d'au moins 1 : 1000. La divulgation concerne également une molécule d'acide nucléique codant pour l'ADN polymérase recombinée dépendante de l'ADN, un procédé pour synthétiser de l'ADN double brin (ADNdb), un procédé pour obtenir la position d'une rupture simple brin dans une molécule d'ADNdb matrice, et un procédé pour introduire des mutations dans l'ADN de cellules bactériennes ou eucaryotes, ou d'organismes.
