Abstract
(EN) Disclosed is an erythrocyte biomimetic coating for enriching circulating tumor cells (CTCs). By means of dense arrangement of erythrocytes on a substrate, non-specific adsorption of leukocytes on the surface of the substrate is avoided; in addition, the surfaces of the erythrocytes are modified with folic acid (FA) or an antibody so that the erythrocytes can directly target CTCs; the functionalized erythrocyte biomimetic coating can capture the CTCs more firmly and more efficiently by means of a cell membrane surface thereof having the fluidity, the capture efficiency is up to 90% or more, and the capture purity of the CTCs is 80% or more. The erythrocytes are derived from a same donor, so that introduction of foreign matters is avoided, and high-purity CTCs can be separated by using an erythrocyte lysis solution.
(FR) Un revêtement biomimétique d'érythrocytes pour enrichir des cellules tumorales circulantes (CTC) est divulgué. Au moyen d'un agencement dense d'érythrocytes sur un substrat, une adsorption non spécifique de leucocytes sur la surface du substrat est évitée; en outre, les surfaces des érythrocytes sont modifiées avec de l'acide folique ou un anticorps de telle sorte que les érythrocytes peuvent directement cibler des CTC; le revêtement biomimétique d'érythrocytes fonctionnalisé peut capturer les CTC plus fermement et plus efficacement au moyen d'une surface de membrane cellulaire correspondante ayant une fluidité, l'efficacité de capture est supérieure ou égale à 90 % et la pureté de capture des CTC est supérieure ou égale à 80 %. Les érythrocytes sont dérivés d'un même donneur, de telle sorte que l'introduction de matières étrangères est évitée, et des CTC de haute pureté peuvent être séparées par l'utilisation d'une solution de lyse d'érythrocytes.
(ZH) 本发明公开了一种富集循环肿瘤细胞的红细胞仿生涂层。通过红细胞在基底上的致密排布,避免白细胞在基底表面非特异性吸附,同时红细胞表面修饰有叶酸(FA)或抗体可直接靶向CTCs,所述功能化红细胞仿生涂层凭借其具有流动性的细胞膜表面,可以实现更牢固更高效地捕获循环肿瘤细胞,捕获效率最高达到了90%以上,CTCs俘获纯度实现80%以上。红细胞源自同一供体,避免了异物引入,利用红细胞裂解液即可分离得到高纯度的CTCs。
