Abstract
(EN) The present invention presents an improved method for identification of neoepitopes useful in active immunotherapy targeting malignant neoplasms. The method integrates identification of somatic variants of expression product with a balanced evaluation of such variants' 1) ability to bind MHC, 2) ability to induce immune responses, 3) clonal coverage in the tumour tissue, and 4) ability to evade immune responses. Also, the method is complemented by a method for purposive deselection of neoepitopes that could induced undesired immune response against normal cells. Also disclosed is a method for preparing immunogenic compositions, a method for treatment of cancer, and a computer system for identifying neoepitopes and neopeptides.
(FR) La présente invention présente une méthode améliorée d'identification de néo-épitopes utiles dans une immunothérapie active ciblant des néoplasmes malins. La méthode intègre l'identification de variants somatiques de produit d'expression par une évaluation équilibrée de 1) leur capacité à se lier au CMH, 2) leur capacité à induire des réponses immunitaires, 3) leur couverture clonale dans le tissu tumoral, et 4) leur capacité à éviter des réponses immunitaires. En outre, la méthode est complétée par une méthode de désélection intentionnelle de néo-épitopes qui pourraient induire une réponse immunitaire indésirable contre des cellules normales. L'invention concerne également une méthode de préparation de compositions immunogènes, une méthode de traitement du cancer, et un système informatique pour identifier des néo-épitopes et des néopeptides.
