Abstract
(EN) A fluorescent cross-linked RNase H mutant conjugate, a miRNA combination and an application thereof. The fluorescent cross-linked RNase H mutant conjugate (i) is as represented by RNase Hv-(Lx-SH-F)n or (ii) comprises an RNase Hv-Lx-ligand and receptor-F, wherein the ligand can bind to the receptor; RNase Hv is an RNase H mutant, which can bind to RNA or RNA-DNA hybrid strands, but cannot cleave RNA; L is a linker, and x is 1-10; SH is an amino acid containing a sulfhydryl group; F is a luminescent functional group, and n is 1-7. The fluorescent cross-linked RNase H mutant conjugate can directly recognize DNA/RNA hybrid strands and can be converted to generate detectable signals without PCR amplification, and can be applied to sensitively and quickly detect RNA.
(FR) L'invention concerne un conjugué mutant de RNase H à réticulation fluorescente, une combinaison de miARN et une application associée. Le conjugué mutant de RNase H à réticulation fluorescente (i) est tel que représenté par RNase Hv-(Lx-SH-F)n ou (ii) comprend un ligand RNase Hv-Lx et un récepteur-F, le ligand pouvant se lier au récepteur; RNase Hv est un mutant de RNase H, pouvant se lier à des brins hybrides d'ARN ou d'ARN-ADN, mais ne pouvant pas cliver l'ARN; L est un coupleur et x représente 1-10; SH est un acide aminé contenant un groupe sulfhydryle; F est un groupe fonctionnel luminescent et n est compris entre 1 et 7. Le conjugué mutant de RNase H à réticulation fluorescente peut reconnaître directement des brins hybrides d'ADN/ARN et peut être converti pour générer des signaux détectables sans amplification par PCR, et peut être appliqué pour détecter de façon sensible et rapide l'ARN.
(ZH) 一种荧光交联的RNase H突变体缀合物、miRNA组合及其应用。所述荧光交联的RNase H突变体缀合物,所述RNase H突变体缀合物(i)如RNase Hv-(Lx-SH-F)n所示,或(ii)包括RNase Hv-Lx-配体、受体-F,所述配体可结合所述受体;其中RNase Hv为RNase H突变体,其能结合RNA或RNA-DNA杂合链,但无法剪切RNA;其中,L为连接子,x为1-10;SH为含有巯基的氨基酸;F为发光官能团,n为1-7。所述荧光交联的RNase H突变体缀合物可直接识别DNA/RNA杂合链并能转换产生可检测信号且无需PCR扩增,可应用于灵敏、快捷地检测RNA。
