Abstract
(EN) Provided are a taxadiene synthase TcTS2, a encoding nucleotide sequence and use thereof. The amino acid sequence of TcTS2 comprises or consists of (a) an amino acid sequence set forth in SEQ ID NO: 1; or (b) a functional homologous sequence having at least 80% sequence similarity with the amino acid sequence set forth in SEQ ID NO: 1; or (c) an amino acid sequence having TcTS2 activity with addition, deletion, or substitution of one or more amino acids to the amino acid sequence set forth in SEQ ID NO: 1. The TcTS2 and the nucleotide sequence encoding the TcTS2 provide new gene resources for improving the yield of paclitaxel, can be used for modifying chassis hosts by means of plant genetic engineering and metabolic engineering strategies and producing paclitaxel and intermediates and the like thereof, and have significant economic and social value.
(FR) L'invention concerne une taxadiène synthase TcTS2, une séquence nucléotidique codante et son utilisation. La séquence d'acides aminés de TcTS2 comprend ou consiste en (a) une séquence d'acides aminés représentée dans SEQ ID NO : 1; ou (b) une séquence homologue fonctionnelle ayant au moins 80 % de similarité de séquence avec la séquence d'acides aminés représentée dans SEQ ID NO : 1; ou (c) une séquence d'acides aminés ayant une activité TcTS2 avec addition, délétion ou substitution d'un ou plusieurs acides aminés à la séquence d'acides aminés représentée dans SEQ ID NO : 1. Le TcTS2 et la séquence nucléotidique codant pour le TcTS2 fournissent de nouvelles ressources génétiques pour améliorer le rendement en paclitaxel, peuvent être utilisés pour modifier des hôtes châssis au moyen de stratégies de génie génétique végétal et de génie métabolique et produire du paclitaxel et des intermédiaires et similaires, et ont une valeur économique et sociale significative.
