Abstract
(EN) The present invention relates to a method for detecting Sarbecovirus which is one of the subgenera of betacoronavirus and novel coronavirus (severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)) which induces coronavirus disease-19, by means of a reverse transcription real-time polymerase chain reaction and, more particularly, to a PNA probe and primer for simultaneously detecting Sarbecovirus and novel coronavirus, and a method for diagnosing an infection by using same. The present invention exhibits higher sensitivity and specificity compared to an existing PCR and RT-PCR method using a DNA probe for coronavirus disease-19 diagnosis, and can amplify and distinguish ORF1ab gene, S gene and N gene of the novel coronavirus and E gene of Sarbecovirus, and thus can analyze an infection of the novel coronavirus and Sarbecovirus with high sensitivity and accuracy.
(FR) La présente invention porte sur une méthode de détection du sarbecovirus qui représente l'un des sous-genres de bêtacoronavirus et du nouveau coronavirus (coronavirus 2 responsable du syndrome respiratoire aigu sévère (SARS-CoV-2)) qui induit la maladie à coronavirus-19, au moyen d'une réaction d'amplification en chaîne par polymérase en temps réel à transcription inverse et, plus particulièrement, une sonde PNA et une amorce pour la détection simultanée du sarbecovirus et du nouveau coronavirus, et une méthode de diagnostic d'une infection faisant appel à celle-ci. La présente invention présente une sensibilité et une spécificité supérieures comparativement à une méthode de PCR et de RT-PCR existante faisant appel à une sonde d'ADN pour le diagnostic de la maladie à coronavirus-19, et peut amplifier et distinguer le gène ORF1ab, le gène S et le gène N du nouveau coronavirus et le gène E du sarbecovirus, et peut ainsi analyser une infection à nouveau coronavirus et à sarbecovirus avec une sensibilité et une précision élevées.
(KO) 본 발명은 역전사 중합효소 연쇄반응(Reverse transcription Real-time polymerase chain reaction)을 이용한 베타코로나바이러스의 하속(subgenus)중 하나인 살베코바이러스(Sarbecovirus) 및 코로나바이러스감염증-19을 일으키는 신종 코로나바이러스(Severe acute respiratory syndrome coronavirus 2, SARS-CoV-2)를 검출하는 방법에 관한 것으로, 더욱 자세하게 살베코바이러스 및 신종 코로나바이러스의 동시 검출용 PNA 프로브와 프라이머 및 이를 이용한 감염여부 판별방법에 관한 것으로, 본 발명에 따르면, 종래 코로나바이러스감염증-19 진단에 사용되는 DNA 프로브를 이용한 PCR과 RT-PCR 방법에 비해 민감도와 특이도가 높으며 신종 코로나바이러스의 ORF1ab 유전자, S 유전자, N 유전자 및 살베코바이러스의 E 유전자를 동시 증폭 및 판별할 수 있어, 고민감도와 높은 정확성을 가지고 신종 코로나바이러스 및 살베코바이러스의 감염 여부분석 가능하다.
