Abstract
(EN) The present invention provides a method for producing a cartilage tissue, said method being characterized by comprising: a step for preparing a cell aggregate production substrate wherein, on a substrate capable of suppressing cell adhesion, a plurality of spots, said spots being formed of a copolymer that contains repeating units derived from monomers represented by formulae (I) and (II) [in the formulae, Ua1, Ua, Ra1, Ra2 and Rb are each as mentioned in the description and claims] are provided; a step for inoculating the substrate with PRRX1 protein-positive human cartilage precursor cells derived from pluripotent stem cells; a step for culturing the cells to form a cell aggregate; and a step for culturing the aggregate to form a cartilage tissue.
(FR) La présente invention concerne un procédé pour la production d'un tissu cartilagineux, ledit procédé étant caractérisé en ce qu'il comprend : une étape consistant à préparer un substrat de production d'agrégat cellulaire dans laquelle, sur un substrat permettant de supprimer l'adhérence cellulaire, plusieurs points sont disposés, lesdits points étant formés d'un copolymère qui contient des unités de répétition dérivées de monomères représentés par les formules (I) et (II) [dans les formules, Ua1, Ua, Ra1, Ra2 et Rb sont chacun tels que mentionnés dans la description et les revendications]; une étape consistant à inoculer le substrat avec des cellules précurseurs de cartilage humain positives à la protéine PRRX1 dérivées de cellules souches pluripotentes; une étape consistant à cultiver les cellules pour former un agrégat cellulaire; et une étape consistant à cultiver l'agrégat pour former un tissu cartilagineux.
(JA) 本発明は、細胞の付着抑制能を有する基板上に、下記式(I)及び(II): [式中、Ua1、U、Ra1、Ra2及びRは、明細書及び特許請求の範囲に記載のとおりである]で表されるモノマーから誘導される繰り返し単位を含む共重合体からなる複数のスポットを備える細胞凝集塊製造用基板を提供する工程;該基板に、多能性幹細胞由来のPRRX1タンパク質陽性であるヒト軟骨前駆細胞を播種する工程;該細胞を培養し細胞凝集塊を作成する工程;該凝集塊を培養し軟骨組織体を作成する工程、を含むことを特徴とする、軟骨組織体の製造方法を提供する。
