Abstract
(EN) The present invention provides an in vitro method for the manufacture of a dendritic cell (DC) cancer vaccine, said method comprising the steps of: (i) providing a plurality of phagocytosable particles, wherein each phagocytosable particle comprises a core and an antigenic construct tightly associated to the core, wherein the antigenic construct comprises at least one epitope peptide having an amino acid sequence corresponding to an amino acid sequence of a part of a protein or peptide known or suspected to be expressed by a cancer cell in a subject; (ii) providing a sample of DCs; and (iii) contacting the sample of DCs with the plurality of phagocytosable particles in vitro and under conditions allowing for the phagocytosis of at least one phagocytosable particle by a DC. The present invention also provides a DC cancer vaccine produced by the method of the invention, and the use a DC cancer vaccine of the invention as a medicament and for the ex vivo expansion of anticancer T-cells.
(FR) La présente invention concerne une méthode in vitro de fabrication d'un vaccin anticancéreux à base de cellules dendritiques (CD), ladite méthode comprenant les étapes consistant : (i) à fournir une pluralité de particules phagocytosables, chaque particule phagocytosable comprenant un noyau et une construction antigénique étroitement associée au noyau, la construction antigénique comprenant au moins un peptide épitope ayant une séquence d'acides aminés correspondant à une séquence d'acides aminés d'une partie d'une protéine ou d'un peptide connu ou suspecté d'être exprimé par une cellule cancéreuse chez un sujet ; (ii) à fournir un échantillon de CD ; et (iii) à mettre en contact l'échantillon de CD avec la pluralité de particules phagocytosables in vitro et dans des conditions permettant la phagocytose d'au moins une particule phagocytosable par une CD. La présente invention concerne également un vaccin anticancéreux à base de CD produit par la méthode selon l'invention, et l'utilisation d'un vaccin anticancéreux à base de CD selon l'invention en tant que médicament et pour l'expansion ex vivo de lymphocytes T anticancéreux.
