Abstract
(EN) A method for determining the type of a nucleotide on a nucleic acid sequence to be analyzed, and a nucleic acid sequencing method. In the method, at least one modified nucleotide for nucleic acid synthesis is separated from a nucleic acid sequence to be analyzed and other components on both sides of a membrane; when the modified nucleotide is transferred to the other side of the membrane under the action of an electric field by means of a nanopore embedded on the membrane, a synthesis reaction is conducted; moreover, the type of a base of the nucleotide is determined according to the change of the electrical properties of the nanopore in the process of the modified nucleotide is transferred by the nanopore, so as to implement sequencing.
(FR) Procédé pour déterminer le type d'un nucléotide sur une séquence d'acide nucléique à analyser, et procédé de séquençage d'acide nucléique. Dans le procédé, au moins un nucléotide modifié pour la synthèse d'un acide nucléique est séparé d'une séquence d'acide nucléique à analyser et d'autres composants des deux côtés d'une membrane ; lorsque le nucléotide modifié est transféré de l'autre côté de la membrane sous l'action d'un champ électrique au moyen d'un nanopore incorporé à la membrane, une réaction de synthèse est réalisée ; de plus, le type d'une base du nucléotide est déterminé selon le changement des propriétés électriques du nanopore dans le processus de transfert du nucléotide modifié par le nanopore, de façon à mettre en œuvre le séquençage.
(ZH) 一种确定待分析核酸序列上核苷酸类型的方法,以及核酸测序的方法。在所述方法中将至少一种用于核酸合成的经修饰的核苷酸与待分析核酸序列以及其他组分分隔于膜的两侧,在电场的作用下经修饰的核苷酸通过嵌在膜上的纳米孔被转运到膜的另一侧时进行合成反应,同时根据经修饰的核苷酸被纳米孔转运的过程中引起纳米孔电学性质的变化来确定核苷酸的碱基类型从而实现测序。
