The invention provides methods of detecting substantially all types of cell free DNA (cfDNA) in biological samples, including nucleosome- bound cfDNA, exosome-bound cfDNA and unbound cfDNA (including double stranded DNA (dsDNA), single stranded DNA (ssDNA) and oligonucleotides), for diagnosis, monitoring and treatment of diseases caused by, or correlated with, increased levels of cfDNA. The method is based on the isolation of cfDNA from body fluid samples by contacting the body fluid with a n affinity agent comprising a linker histone, e.g. Hl-3.

L'invention concerne des procédés de détection de sensiblement tous les types d'ADN acellulaire (cfDNA) dans des échantillons biologiques, comprenant de l'ADN acellulaire lié à un nucléosome, de l'ADN acellulaire lié à un exosome et de l'ADN acellulaire non lié (y compris de l'ADN double brin (ADNdb), un ADN simple brin (ADNsb) et des oligonucléotides), pour le diagnostic, la surveillance et le traitement de maladies provoquées par des taux accrus d'ADN acellulaire, ou leur étant corrélées. Le procédé est basé sur l'isolement d'ADN acellulaire à partir d'échantillons de fluide corporel par mise en contact du fluide corporel avec un agent d'affinité n comprenant une histone de liaison, par exemple h1-3.
