Abstract

The present disclosure provides methods to recover repertoires of T cell receptors (TCRs). In some embodiments, TCR repertoires are recovered from non-viable samples. In some embodiments, libraries of TCRaβ pairs are created. In some embodiments, the methods disclosed are used for cancer immunotherapy or diagnostic purposes. Described herein, in some embodiments, are methods of identifying nucleotide sequences encoding T cell receptor a (TCRα)- and TCRβ-chains from a combinatorial library of nucleic acids.

La présente invention concerne des procédés permettant de récupérer des répertoires de récepteurs de lymphocytes T (TCR). Dans certains modes de réalisation, les répertoires de TCR sont récupérés à partir d'échantillons non viables. Dans certains modes de réalisation, des bibliothèques de paires de TCRaβ sont créées. Dans certains modes de réalisation, les méthodes selon l'invention sont utilisées pour une immunothérapie anticancéreuse ou à des fins de diagnostic. Dans certains modes de réalisation, l'invention concerne des procédés d'identification de séquences nucléotidiques codant pour des chaînes TCRα (récepteur a des lymphocytes T) et TCRβ à partir d'une bibliothèque combinatoire d'acides nucléiques.
