Abstract

The present invention relates to a method for modifying a target site in a host cell comprising contacting said host cell with a Cas nuclease, wherein said host cell is further contacted with a low-affinity Cas inhibitor and/or a sub-inhibitory concentration of a Cas inhibitor; and to a method for improving specificity of a Cas nuclease, comprising a) providing a Cas nuclease; and b) contacting said Cas nuclease with a low-affinity Cas inhibitor or a sub-inhibitory concentration of a Cas inhibitor; and c) thereby improving specificity of said Cas enzyme. Further, the present invention relates to compositions, polypeptides, uses and methods related thereto.

La présente invention concerne un procédé de modification d'un site cible dans une cellule hôte comprenant la mise en contact de ladite cellule hôte avec une nucléase Cas, ladite cellule hôte étant en outre mise en contact avec un inhibiteur de Cas de faible affinité et/ou une concentration sous-inhibitrice d'un inhibiteur de Cas; et un procédé d'amélioration de la spécificité d'une nucléase Cas, consistant à a) fournir une nucléase Cas ; et b) mettre en contact ladite nucléase Cas avec un inhibiteur de Cas de faible affinité ou une concentration sous-inhibitrice d'un inhibiteur de Cas ; et c) améliorer ainsi la spécificité de ladite enzyme Cas. En outre, la présente invention concerne des compositions, des polypeptides, des utilisations et des procédés associés.
