Abstract
(EN)
Compositions and methods for amplifying RNA by replication using transcription polymerases are disclosed. Such replicated RNAs can be used in various applications such as RNAi therapeutics, diagnostic probes, RNA sequencing, directed evolution of RNA aptamers without intermediate conversion to DNA, and RNA vaccines. The transcription polymerases comprise T7 bacteriophage RNA polymerase.

L'invention concerne des compositions et des procédés permettant l'amplification d'ARN par réplication à l'aide de polymérases de transcription. De tels ARN répliqués peuvent être utilisés dans diverses applications telles que les agents thérapeutiques à base d'ARNi, les sondes de diagnostic, le séquençage d'ARN, l'évolution dirigée d'aptamères d'ARN sans conversion intermédiaire en ADN, et les vaccins à ARN. Parmi lesdites polymérases de transcription figurent l'ARN polymérase du bactériophage T7.
