The present disclosure describes systems and methods to make predictions classifying one or more properties of a binding protein such as an antibody, for example, antibody affinity or specificity for an antigen. The system can include one or more machine learning models that can extrapolate complex relationships between amino acid sequence and function. The system can be trained on high-quality training data generated through a two-step single-site and combinatorial deep mutational scanning approach. The trained models can then make predictions on novel variant sequences generated in silico. The present disclosure describes amino acid sequences generated by the systems and methods provided, and uses of the generated sequences to produce proteins for therapeutic and diagnostic use.

La présente invention concerne des systèmes et des procédés pour effectuer des prédictions en classant une ou plusieurs propriétés d'une protéine de liaison telle qu'un anticorps, par exemple, une affinité ou une spécificité d'un anticorps pour un antigène. Le système peut comprendre un ou plusieurs modèles d'apprentissage automatique qui peuvent extrapoler des relations complexes entre la séquence d'acides aminés et une fonction. Le système peut être entraîné sur des données d'apprentissage de haute qualité générées par un seul site en deux étapes et une approche de balayage de mutation profonde combinatoire. Les modèles entraînés peuvent ensuite effectuer des prédictions sur de nouvelles séquences variantes générées in silico. La présente invention concerne des séquences d'acides aminés générées par les systèmes et les procédés décrits, et des utilisations des séquences générées pour produire des protéines à usage thérapeutique et diagnostique.
