The purpose of the present invention is to provide a humanized mouse that enables the long-term survival of human hematopoietic stem cells. The present invention is a humanized rodent in which human neutrophils circulate peripherally, and is obtained by transplanting human hematopoietic cells into a human G-CSF gene knocked-in rodent that expresses human G-CSF and does not express rodent G-CSF receptor. The human G-CSF gene knocked-in rodent is an immunodeficient rodent in which a human G-CSF gene has been knocked into the G-CSF receptor gene locus and the function of the G-CSF receptor has been lost.

Le but de la présente invention est de fournir une souris humanisée qui permet la survie à long terme de cellules souches hématopoïétiques humaines. La présente invention est un rongeur humanisé dans lequel des neutrophiles humains circulent de manière périphérique, et qui est obtenu par transplantation de cellules hématopoïétiques humaines dans un rongeur possédant le gène humain G-CSF activé exprimant le G-CSF humain et n'exprimant pas le récepteur de G-CSF de rongeur. Le rongeur possédant le gène humain G-CSF activé est un rongeur immunodéficient dans lequel un gène G-CSF humain a été inactivé dans le locus du gène du récepteur G-CSF et la fonction du récepteur G-CSF a été perdue.

本発明は、ヒト造血幹細胞の長期生着を可能とするヒト化マウスの提供を目的とする。本発明はG-CSFレセプター遺伝子座にヒトG-CSF遺伝子をノックインし、G-CSFレセプターの機能を欠損した免疫不全げっ歯類動物であって、ヒトG-CSFが発現しげっ歯類のG-CSFレセプターを発現しない、ヒトG-CSF遺伝子ノックインげっ歯類動物にヒト造血幹細胞を移植して得られる、ヒト好中球が末梢を循環しているヒト化げっ歯類動物である。
