The present invention relates to an in vitro method for diagnosing, stratifying, prognosing and/or monitoring a lung disease and/or a respiratory disease, as well as an in vitro method for assessing the effectiveness of a treatment for a lung disease and/or a respiratory disease, comprising determining or measuring the value Anl of at least one rheological parameter on a mucous sample A of a subject, in a non-linear zone of the deformation curve of the sample, and determining or measuring the value Al of at least one rheological parameter on said mucous sample A, in a linear zone of the deformation curve of the sample, wherein the rheological parameter Anl comprises at least the critical constraint (σc) and the rheological parameter Al comprises at least the complex modulus G*.
