Abstract

A method for identifying individual female sea cucumbers having ovaries in development for Holothuria scabra, comprising: extracting pseudocoelom fluid of a Holothuria scabra; performing centrifugal separation to separate out vitellin; performing SDS-PAGE electrophoresis and dyeing; on the basis of the electrophoresis result, distinguishing female sea cucumbers having ovaries in development, male sea cucumbers, and Holothuria scabras having undeveloped reproductive glands. Samples of individual female sea cucumbers having ovaries in development exhibit characteristic bands of 200 to 250kD, and samples of both individuals having undeveloped reproductive glands and individual male sea cucumbers exhibit no characteristic bands. Individual female sea cucumbers having ovaries in development can be identified by extracting pseudocoelom fluid samples while not harming Holothuria scabras, providing application values to the selection of parent sea cucumbers during artificial seedling development.

L'invention concerne un procédé d'identification d'holothuries femelles individuelles ayant des ovaires en développement de l'holothuria scabra, comprenant : l'extraction d'un fluide pseudocoelom d'une holothuria scabra ; la réalisation d'une séparation centrifuge pour séparer la viteline ; la réalisation d'une électrophorèse SDS-PAGE et la teinture ; sur la base du résultat d'électrophorèse, la distinction entre les holothuries femelles ayant des ovaires en développement, les holothuries mâles et l'espèce holothuria scabra ayant des glandes reproductrices non développées. Des échantillons d'holothuries femelles individuelles ayant des ovaires en développement présentent des bandes caractéristiques de 200 à 250 kD tandis que des échantillons aussi bien des individus ayant des glandes de reproduction non développées que des holothuries mâles individuelles ne présentent pas de bandes caractéristiques. Il est possible d'identifier des holothuries femelles individuelles ayant des ovaires en développement en extrayant des échantillons de fluide de pseudocoelom sans nuire à l'holothuria scabra, en fournissant des valeurs d'application à la sélection d'holothuries parents pendant le développement de semis artificiel.

一种鉴别糙海参卵巢发育期雌参个体的方法,包括:抽取糙海参的体腔液,然后离心分离出卵黄蛋白,再经SDS-PAGE电泳和染色,通过电泳结果将卵巢发育期的雌参、雄参以及性腺未发育的糙海参区别开来。卵巢发育期雌参个体样品呈现200-250kD的特征条带,而性腺未发育个体和雄参个体样品均无出现该特征条带。可以在不损伤糙海参的情况下,通过抽取体腔液样品鉴别卵巢发育期雌参个体,对于人工育苗时亲参的选取具有应用价值。
