Abstract

A method of differentiating pluripotent stem cells into hemangioblasts comprising incubating the pluripotent stem cells in a first serum-free differentiation medium comprising bone morphogenetic protein 4 (BMP4), vascular endothelial growth factor (VEGF) and stem cell factor (SCF) to induce differentiation of the pluripotent stem cells into hemangioblasts or hemangioblast-containing embryoid bodies is provided. The hemangioblasts or embryoid bodies may be cultured in a second differentiation medium comprising at least granulocyte-macrophage colony stimulating factor (GM-CSF), macrophage colony stimulating factor (M-CSF) and interleukin-3 (IL-3) for a period of time sufficient to generate alveolar-like macrophages.

Procédé de différenciation de cellules souches pluripotentes en hémangioblastes comprenant l'incubation des cellules souches pluripotentes dans un premier milieu de différenciation sans sérum comprenant la protéine morphogénétique osseuse 4 (BMP4), le facteur de croissance vasculaire endothéliale (VEGF) et Le facteur des cellules souches (SCF) pour induire la différenciation des cellules souches pluripotentes en hémangioblastes ou en corps embryoïdes contenant des hémangioblastes. Les hémangioblastes ou les corps embryoïdes peuvent être cultivés dans un second milieu de différenciation comprenant au moins un facteur stimulant les colonies de granulocytes-macrophages (GM-CSF), le facteur de stimulation des colonies de macrophages (M-CSF) et l'interleukine-3 (IL-3) pendant une durée suffisante pour produire des macrophages de type alvéolaire.
