A chip and an operating method therefor, and a detection device. The chip (100) comprises a detection cavity (110) and a working electrode (120). The detection cavity (110) is configured to accommodate a plurality of droplets. The working electrode (120) is provided in the detection cavity (110) and configured to regularly arrange the plurality of droplets in the detection cavity (110) along the extension direction of the working electrode (120). The chip (100) can make the droplets arranged regularly, thereby implementing the three-dimensional control of droplet positions, reducing the complexity and costs of an optical system used for detection, decreasing the difficulty of image processing, improving the detection accuracy and detection efficiency, achieving the miniaturization and integration of detection chips, and reducing manual operations.
