Abstract

Provided are u-PA polypeptides and fusion proteins containing the u-PA polypeptides. The u-PA polypeptides are modified to have altered activity and/or specificity so that they cleave a complement protein, such as complement protein C3, to thereby inhibit complement activation. The modified u-PA polypeptides and fusion proteins that inhibit complement activation can be used for treatment of diseases and conditions that are mediated by complement activation, or in which complement activation plays a role. These disorders include ischemic and reperfusion disorders, including myocardial infarction and stroke, sepsis, autoimmune diseases, diabetic retinopathies, age-related macular degeneration, transplanted organ rejection, inflammatory diseases and diseases with an inflammatory component.

L'invention concerne des polypeptides u-PA et des protéines de fusion contenant les polypeptides u-PA. Les polypeptides u-PA sont modifiés pour présenter une activité et/ou une spécificité modifiées de façon à cliver une protéine du complément, telle que la protéine C3 du complément, pour inhiber ainsi l'activation du complément. Les polypeptides u-PA-1 modifiés et les protéines de fusion qui inhibent l'activation du complément peuvent être utilisés pour le traitement de maladies et d'affections médiées par l'activation du complément, ou dans lesquelles l'activation du complément joue un rôle. Ces troubles comprennent des troubles ischémiques et de reperfusion, notamment l'infarctus du myocarde et l'accident vasculaire cérébral, le sepsis, les maladies auto-immunes, les rétinopathies diabétiques, la dégénérescence maculaire liée à l'âge, le rejet d'organe transplanté, les maladies inflammatoires et les maladies avec un composant inflammatoire.
