The present invention generally relates to a process for the preparation of plecanatide. Specifically, the present invention relates to an improved process for preparation of plecanatide using a non-linear solid phase peptide synthesis. In particular, the process for preparation of plecanatide of the present invention involves solid phase synthesis of peptide fragments of five amino acid units using 2-chlorotrityl chloride (2-ClTrt) resin and eleven amino acid unit using Wang resin.

La présente invention concerne un procédé de préparation de plecanatide. Plus particulièrement, la présente invention concerne un procédé amélioré pour la préparation de plecanatide au moyen d'une synthèse peptidique en phase solide non linéaire. En particulier, le procédé de préparation de plecanatide selon la présente invention implique une synthèse en phase solide de fragments peptidiques de cinq unités d'acides aminés à l'aide de résine de chlorure de 2-chlorotrityle (2-ClTrt) et onze unités d'acides aminés à l'aide de résine Wang.
