Systems and methods for inferring a status of a cell population are provided. Described techniques allow deconvolving a first clonal population comprising a first plurality of cells of a species, wherein nucleic acid sequence reads from each cell in the first plurality of cells are obtained. The nucleic acid sequence reads are mapped into bins representing portions of a reference genome, and a pattern of sequence read counts for each cell across the multiple bins is used to assign a cell to a group, thereby inferring a mitotic status of the cell. The assignment of nucleic acid sequence reads into bins is also be used for segregating cells into classes based on a status of a certain biological marker in each cell. Comparison of sequence read counts for a subset of bins across the cell classes allows evaluating effect of a compound on a cell status.

L'invention concerne des systèmes et des procédés permettant de déduire l'état d'une population cellulaire. Les techniques décrites permettent de d'effectuer la déconvolution d'une première population clonale comprenant une première pluralité de cellules d'une espèce, permettant d'obtenir des relevés de la séquence d'acide nucléique dans chaque cellule dans la première pluralité de cellules. Les relevés de séquence d'acides nucléiques sont mises en correspondance dans des compartiments représentant des parties d'un génome de référence, et un motif de comptes de relevés de séquence pour chaque cellule à travers les multiples compartiments est utilisé pour attribuer une cellule à un groupe, moyennant quoi on peut déduire un état mitotique de la cellule. L'attribution de relevés de séquences d'acides nucléiques dans des compartiments peut également être utilisée pour la ségrégation de cellules en classes sur la base d'un état d'un certain marqueur biologique dans chaque cellule. La comparaison de relevés de lecture de séquence pour un sous-ensemble de compartiments à travers les classes de cellules permet d'évaluer l'effet d'un composé sur un état de cellule.
