The present invention relates to an isolated variant Ire1 polypeptide, comprising (a) an amino acid substitution at position 153 of the Trichoderma reesei Ire1 polypeptide of SEQ ID NO: 2; or (b) an amino acid substitution at a position corresponding to position 153 of the Trichoderma reesei Ire1 of SEQ ID NO: 2 and has at least 70%, but less than 100%, sequence identity to the amino acid sequence of SEQ ID NO: 2. The present invention also relates to a recombinant filamentous fungal host cell comprising the mutated ire1 gene encoding the variant Ire1 polypeptide for producing a secreted heterologous polypeptide of interest, a method for producing the secreted heterologous polypeptide of interest in the recombinant filamentous fungal host cell, and a method of improving the productivity or yield of the secreted heterologous polypeptide of interest in the recombinant filamentous fungal host cell.

La présente invention concerne un polypeptide Ire1 variant isolé, comprenant (a) une substitution d'acide aminé en position 153 du polypeptide Ire1 de Trichoderma Reesei de SEQ ID NO : 2; ou (b) une substitution d'acides aminés en position correspondant à la position 153 de la Trichoderma Reesei Ire1 de SEQ ID NO : 2 et qui présente au moins 70 %, mais moins de 100 %, une identité de séquence avec la séquence d'acides aminés de SEQ ID NO : 2. La présente invention concerne également une cellule hôte fongique filamenteuse de recombinaison comprenant le gène Ire1 muté codant pour le polypeptide Ire1 variant pour produire un polypeptide d'intérêt hétérologue sécrété, un procédé de production du polypeptide d'intérêt hétérologue sécrété dans la cellule hôte fongique filamenteuse de recombinaison, et un procédé permettant d'améliorer la productivité ou le rendement du polypeptide d'intérêt hétérologue sécrété dans la cellule hôte fongique filamenteuse de recombinaison.
