The present invention relates to a Staphylococcus aureus-specific nucleic acid probe and a use thereof. The nuclease secreted from Staphylococcus aureus cleaves the probe of the present invention into a quencher and a fluorophore which are separated from each other distant enough to bring about a significant increase in fluorescent signal, thereby detecting and identifying Staphylococcus aureus as well as confirming susceptibility to antibacterial agents accurately, rapidly, and easily. Therefore, the S. aureus specific FRET probe based AST (SF-AST) system of the present invention can be easily conducted and is fast and economically advantageous. Furthermore, being designed to be suitable for reading on a microtitration plate reader, the system can readily utilize conventional techniques and provides a basis for clinical therapy.

La présente invention concerne une sonde d'acide nucléique spécifique au Staphylococcus aureus et une utilisation de celle-ci. La nucléase sécrétée par le Staphylococcus aureus clive la sonde de la présente invention en un extincteur et un fluorophore séparés l'un de l'autre et suffisamment éloignés de manière à provoquer une augmentation significative du signal fluorescent permettant ainsi de détecter et d'identifier le staphylocoque doré ainsi que la sensibilité à des agents antibactériens de manière précise, rapide et facile. Par conséquent, le système AST (SF-AST) sur base de sonde FRET spécifique au Staphylococcus aureus de la présente invention est facile à réaliser, rapide et économiquement avantageux. En outre, étant conçu pour être approprié pour une lecture sur un lecteur de plaques de microtitration, le système peut facilement utiliser des techniques classiques et fournit une base pour une thérapie clinique.

본 발명은 황색포도상구균-특이 핵산 프로브 및 이의 용도에 관한 것으로, 본 발명의 프로브는 황색포도상구균으로부터 분비되는 뉴클레아제에 의해 절단되어 퀀처(quencher)와 형광단(fluorophore)이 거리적으로 분리되면서 유의한 형광 신호의 증가가 구현되어, 황색포도상구균의 검출 및 동정 뿐만 아니라 항균제 감수성을 정확하고 신속용이하게 확인할 수 있다. 따라서, 본 발명의 황색포도상구균-특이 항생제 내성 진단(SF-AST; S. aureus specific FRET probe based AST) 시스템은 쉽게 실시할 수 있고, 신속하며, 경제적일 뿐만 아니라, 마이크로타이트레이션 플레이트 리더로 판독하기에 적합하도록 설계되어 기존 기술들을 쉽게 활용할 수 있어 임상 치료의 기초를 제공한다.
