The present disclosure provides a method of preparing a hydrogel product comprising crosslinked glycosaminoglycan molecules at a concentration Cfinal (mg/mL). The method comprising the steps of: a) crosslinking a glycosaminoglycan having a molecular weight of above 700 kDa in conditions so as to provide a glycosaminoglycan hydrogel crosslinked by amide bonds and having a Cmin (mg/mL) that is above Cfinal/2, wherein Cmin is the concentration of gel-form glycosaminoglycan in the gel when fully swollen in physiological buffer; b) subjecting the hydrogel from step a) to post-crosslinking degradation of the glycosaminoglycan backbone, thereby reducing the Cmin of the hydrogel to a value that is below Cfinal/2; and c) formulating the partially degraded hydrogel obtained from step b) to form an injectable hydrogel composition having a concentration of glycosaminoglycan molecules of Cfinal (mg/mL).

La présente invention concerne un procédé de préparation d’un produit hydrogel comprenant des molécules de glycosaminoglycane réticulé à une concentration Cfinale (mg/ml). Le procédé comprenant les étapes de : a) réticulation d’un glycosaminoglycane ayant un poids moléculaire supérieur à 700 kDa dans des conditions permettant de fournir un hydrogel de glycosaminoglycane réticulé par des liaisons amides et ayant une Cmin (mg/ml) qui est supérieure à Cfinale/2, où Cmin est la concentration du glycosaminoglycane sous forme de gel dans le gel lorsqu’entièrement gonflé dans du tampon physiologique ; b) la soumission de l’hydrogel de l’étape a) à la dégradation post-réticulation du squelette glycosaminoglycane, ce qui réduit la Cmin de l’hydrogel à une valeur qui est inférieure à Cfinale/2 ; et c) la formulation de l’hydrogel partiellement dégradé obtenu à partir de l’étape b) pour former une composition d’hydrogel injectable ayant une concentration de molécules de glycosaminoglycane de Cfinale (mg/ml).
