Abstract

The present invention relates to a solid-phase chelator material inter alia usable for the purification of proteins. The solid-phase chelator material comprises a solid phase, polyamine groups bound to the solid phase and chelating groups bound to the polyamine groups. At least a part of the polyamine groups is connected with at least two chelating groups per polyamine group. Each chelating group comprises one or several aminopolycarboxylic acid groups (APA groups), with the proviso that the number of APA groups per polyamine group connected with at least two cheating groups is at least three. Several APAs may form linear chelator chains or branched chelator chains, in which the APA groups are interconnected by bifunctional or trifunctional linkers. Metal ion loaded solid-phase chelator materials of the invention show a strong metal binding, a high stability against chelators, reductants and alkaline solutions, a high affinity to proteins and an extremely high binding capacity. Furthermore, the invention relates to a method for producing said solid-phase chelator material and to a method for purifying his-tagged recombinant polypeptides and proteins, phosphoproteins and metal-coordinating proteins with a metal ion loaded solid-phase chelator material.

La présente invention concerne un matériau chélateur en phase solide pouvant être utilisé, entre autres, pour la purification de protéines. Le matériau chélateur en phase solide comprend une phase solide, des groupes polyamine liés à la phase solide et des groupes chélatants liés aux groupes polyamine. Au moins une partie des groupes polyamine est reliée à au moins deux groupes chélatants par groupe polyamine. Chaque groupe chélatant comprend un ou plusieurs groupes acide aminopolycarboxylique (groupes APA), à condition que le nombre de groupes APA par groupe polyamine relié à au moins deux groupes chélatants soit au moins égal à trois. Plusieurs APA peuvent former des chaînes de chélateur linéaires ou des chaînes de chélateur ramifiés, les groupes APA étant inter-reliés par des lieurs bifonctionnels ou trifonctionnels. Les matériaux chélateurs à phase solide chargés d'ions métalliques de l'invention présentent une forte liaison métallique, une haute stabilité contre des chélateurs, des réducteurs et des solutions alcalines, une haute affinité aux protéines et une capacité de liaison extrêmement élevée. En outre, l'invention concerne un procédé de production dudit matériau chélateur en phase solide et un procédé de purification de protéines et de polypeptides recombinants marqués à l'histidine, de phosphoprotéines et de protéines de coordination métallique avec un matériau chélateur en phase solide chargé d'ions métalliques.
