Disclosed is a method for detecting microRNAs based on rolling circle amplification and DNA origami, in particular comprising: amplifying a target signal by means of rolling circle amplification, while introducing an orderly arranged detection unit by means of DNA origami, so as to detect nucleic acid molecules. The method has the advantages that (1) by carrying out signal amplification by means of rolling circle amplification, a high-temperature denaturation process is not needed, and the reaction is quick and the detection sensitivity is improved; and (2) the amplification product is folded by means of DNA origami, so that introduced detection sequences can be arranged in an orderly manner, and signal interference caused by directly surrounding the detection sequences is avoided.

Procédé de détection de micro-ARN sur la base d'une amplification circulaire et d'un origami d'ADN, comprenant en particulier l'amplification d'un signal cible au moyen d'une amplification circulaire, tout en introduisant une unité de détection agencée de manière ordonnée au moyen d'un origami d'ADN, de façon à détecter des molécules d'acide nucléique. Le procédé présente les avantages suivants : (1) en réalisant une amplification de signal au moyen d'une amplification circulaire, un procédé de dénaturation à haute température n'est pas nécessaire, la réaction est rapide et la sensibilité de détection est améliorée; et (2) le produit d'amplification est plié au moyen d'un origami d'ADN, de telle sorte que des séquences de détection introduites peuvent être agencées de manière ordonnée, et un brouillage de signal provoqué par l'environnement direct des séquences de détection est évité.

一种基于滚环扩增和DNA折纸术检测microRNAs的方法,具体涉及一种以滚环扩增放大靶标信号,同时利用DNA折纸术引入并有序排列检测单元,从而实现对核酸分子的检测。优点在于:(1)以滚环扩增进行信号放大,不需要高温变性过程,反应迅速,提高检测灵敏度。(2)利用DNA折纸的技术对扩增产物进行折叠,使引入的检测序列可以有序的排布,避免了检测序列直接环绕造成的信号干扰。
