Provided is a method for producing a peptide-nucleic acid complex that contains a peptide and nucleic acid encoding the peptide. The method comprises a step of preparing a nucleic acid containing a first coding sequence that encodes a peptide, a second coding sequence that encodes a transpeptidase, and a third coding sequence that encodes a motif for recognizing the transpeptidase, the nucleic acid including a transpeptidase N-terminal substrate motif added thereto; a step of synthesizing, from the transpeptidase N-terminal substrate motif-added nucleic acid and using a cell-free protein synthesis system, a chimeric protein that contains a domain of the peptide, a domain of the transpeptidase, and the motif for recognizing the transpeptidase; and a step of forming the peptide-nucleic acid complex by means of the transpeptidase domain.

L'invention concerne un procédé de production d'un complexe peptide-acide nucléique qui contient un peptide et un acide nucléique codant pour le peptide. Le procédé comprend une étape de préparation d'un acide nucléique contenant une première séquence de codage qui code un peptide, une seconde séquence de codage qui code une transpeptidase, et une troisième séquence de codage qui code un motif pour reconnaître la transpeptidase, l'acide nucléique comprenant un motif de substrat de transpeptidase N-terminal ajouté à celui-ci ; une étape de synthèse, à partir de l'acide nucléique ajouté à un motif de substrat de transpeptidase N-terminal et utilisant un système de synthèse de protéines exempt de cellules, une protéine chimérique qui contient un domaine du peptide, un domaine de transpeptidase, et un motif de reconnaissance de la transpeptidase ; et une étape de formation du complexe peptide-acide nucléique au moyen du domaine de transpeptidase.
