Disclosed by the present invention are a transgenic maize nuclear male-sterile maintainer line and an application thereof. The maintainer line is constructed by introducing an exogenous T-DNA vector into a maize genome. Other than a fertility restoring gene having genic male sterility, the introduced exogenous T-DNA also contains at least two of the following three functional fragments: 1) an expression frame for expressing fluorescent protein in seed endosperms; 2) a DNA fragment capable of inhibiting the expression of corn herbicide resistance gene CYP81A9; and 3) a DNA fragment capable of inhibiting the expression of cell wall converting enzyme InCW2 specifically expressed in maize endosperm. The present invention simultaneously utilizes at least two of the three methods for discriminating transgenic and non-transgenic, thereby further reducing errors that may be caused by relying only on one discrimination (or selection) method.

La présente invention concerne une ligne de maintien de la stérilité mâle nucléaire du maïs transgénique et une utilisation associée. La ligne de maintien est construite par introduction d'un vecteur D'ADN-T exogène dans un génome de maïs. En dehors d'un gène de restauration de la fertilité ayant une stérilité mâle génique, l'ADN-T exogène introduit contient également au moins deux des trois fragments fonctionnels suivants : 1) Un cadre d'expression pour exprimer une protéine fluorescente dans des endospermes de graines ; 2) un fragment d'ADN capable d'inhiber l'expression du gène CYP81A9 de résistance aux herbicides de maïs ; et 3) un fragment d'ADN capable d'inhiber l'expression de l'enzyme de conversion de paroi cellulaire InCW2 spécifiquement exprimée dans l'endosperme de maïs. La présente invention utilise simultanément au moins deux des trois procédés pour distinguer des produits transgéniques et non transgéniques, ce qui permet de réduire davantage les erreurs qui peuvent être provoquées par l'utilisation d'un unique procédé de discrimination (ou de sélection).

本发明公开了一种转基因玉米核雄性不育保持系及应用,所述保持系是将外源T-DNA载体导入玉米基因组构建而成,导入的外源T-DNA中除了雄性核不育的育性恢复基因以外,还包含如下3种功能片段中至少2个功能片段:1)在种子胚乳中表达一种荧光蛋白质的表达框;2)一种可以抑制玉米抗除草剂基因CYP81A9表达的DNA片段;3)一种可以抑制玉米胚乳特异表达的细胞壁转化酶InCW2表达的DNA片段。本发明同时利用上述三种区分转基因与非转基因方法中至少2种方法,进一步降低仅仅依靠一种区分(或选择)方法可能导致的错误。
