Abstract

A novel PTH(1-34) mimetic peptide based on protein domain reconstruction. The mimetic peptide contains peptide fragment repeats at the 29-34th positions, and the 34th position of the mimetic peptide and the last amino acid of the peptide fragment repeats at the 29-34th positions are changed from phenylalanine (Phe, F) into tyrosine (Tyr, Y), so that MY-3 peptide is formed. Preclinical experiments show that the MY-3 mimetic peptide may promote osteoblast differentiation and has a very good effect in treating osteoporosis and promoting bone growth. The anti-osteoporosis effect of the MY-3 mimetic peptide is better than that of the PTH (1-34), so that the MY-3 mimetic peptide has a significant clinical application prospect.

L'invention concerne un nouveau peptide mimétique de PTH(1-34) basé sur la reconstruction de domaine protéique. Le peptide mimétique contient des répétitions de fragments peptidiques aux positions 29 à 34, et la position 34 du peptide mimétique et le dernier acide aminé des répétitions de fragments peptidiques aux positions 29 à 34 sont modifiées de phénylalanine (Phe, F) en tyrosine (Tyr, Y), de sorte que le peptide MY-3 soit formé. Des expériences précliniques montrent que le peptide mimétique MY-3 peut favoriser la différenciation des ostéoblastes et a un très bon effet dans le traitement de l'ostéoporose et la promotion de la croissance osseuse. L'effet anti-ostéoporose du peptide mimétique MY-3 est meilleur que celui de la PTH (1-34), de sorte que le peptide mimétique MY-3 présente une perspective d'application clinique significative.
