Disclosed are methods and compositions for real-time monitoring cellular calcium ion dynamics by red-shifted genetically encoded indicators under optogenetic control, in which millisecond-timescale of temporal control of optical activation or inactivation and signal recording can be achieved. The methods include artifact-free functional imaging in conjunction with optogenetic tools for studying cellular physiology, signal transduction and neuronal activity. Thus, all-optical and non-invasive approaches for drug screening, toxicity testing and assessment of cell functions may be provided.

L'invention concerne des procédés et des compositions pour la surveillance en temps réel de la dynamique des ions calcium cellulaires par des indicateurs génétiquement codés décalés vers le rouge sous une commande optogénétique, dans lequel il est possible de contrôler en quelques millisecondes l'activation ou l'inactivation optique et l'enregistrement de signaux. Les procédés comprennent une imagerie fonctionnelle sans artefact en conjonction avec des outils optogénétiques pour étudier la physiologie cellulaire, la transduction de signal et l'activité neuronale. Ainsi, toutes les approches non-optiques et non invasives pour le criblage de médicaments, le test de toxicité et l'évaluation de fonctions cellulaires peuvent être fournies.
