The present disclosure is directed to dipeptide repeat (DPR) peptide immunogen constructs, compositions containing the constructs, antibodies elicited by the constructs, and methods for making and using the constructs and compositions thereof. The disclosed DPR peptide immunogen constructs contain a B cell epitope derived from a DPR protein from C9orf72, including repeats of poly-GA, poly-GP, poly-GR, poly-PR, and poly-PA, linked to a heterologous T helper cell (Th) epitope directly or through an optional heterologous spacer. The B cell epitope portion of the peptide immunogen constructs contain about 10 to about 25 repeats of the respective dipeptide sequence. The disclosed peptide immunogen constructs stimulate the generation of highly specific antibodies directed against the DPR sequences. The disclosed peptide immunogen constructs can be used as an immunotherapy for patients suffering from amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), and/or any other condition caused by the presence of DPRs.

La présente invention concerne des constructions immunogènes peptidiques dirigées contre des répétitions dipeptidiques (DPR), des compositions contenant les constructions, des anticorps produits par les constructions, et des procédés de préparation et d'utilisation des constructions et des compositions en contenant. Les constructions immunogènes peptidiques DPR décrites contiennent un épitope de lymphocyte B dérivé d'une protéine DPR provenant de C9orf72, comprenant des répétitions de poly-GA, poly-GP, poly-GR, poly-PR, et poly-PA, liés à un épitope de lymphocyte T auxiliaire hétérologue (Th) directement ou par l'intermédiaire d'un espaceur hétérologue facultatif. La partie épitope de lymphocyte B des constructions immunogènes peptidiques contient environ 10 à environ 25 répétitions de la séquence dipeptidique respective. Les constructions immunogènes peptidiques décrites stimulent la génération d'anticorps hautement spécifiques dirigés contre les séquences DPR. Les constructions immunogènes peptidiques décrites peuvent être utilisées comme une immunothérapie pour des patients souffrant de sclérose latérale amyotrophique (SLA), de démence frontotemporale (FTD), et/ou de toute autre condition provoquée par la présence de DPR.
