Disclosed is a method for preparation of exosomes having a hair regeneration effect, the method comprising the steps of: providing direct or indirect ultrasonic stimuli for cells; incubating a mixture of the cells and a medium for a predetermined period of time; and separating exosomes from the mixture, wherein the providing of direct ultrasonic stimuli refers to applying ultrasonic stimuli to a medium containing cells and the providing of indirect ultrasonic stimuli refers to applying ultrasonic stimuli to a cell-free medium which is then mixed with the cell. Such a method for preparation of exosomes allows the acquirement of exosomes having a hair regeneration effect at high yield within a short period of time through the simple process of ultrasonication from stem cells difficult to handle as well as somatic cells that can be easily obtained and maintained. The exosomes thus obtained also contain a large quantity of various factors. The exosomes and a composition containing same have increased numbers of various factors capable of inducing hair regeneration. Having a phospholipid-based membrane structure, the exosomes can easily penetrate into the scalp and deliver a material at a high rate to effectively induce hair regeneration. The exosomes are advantageously free of the adverse effects resulting from the use of synthetic medications.

L'invention concerne un procédé de préparation d'exosomes possédant un effet de régénération sur les cheveux, le procédé comprenant les étapes consistant : à fournir des stimuli ultrasonores directs ou indirects destinés aux cellules ; à incuber un mélange comprenant les cellules et un milieu pendant une période de temps prédéterminée ; et à séparer les exosomes du mélange, la fourniture de stimuli ultrasonores directs se rapportant à l'application de stimuli ultrasonores à un milieu contenant des cellules, et la fourniture de stimuli ultrasonores indirects se rapportant à l'application de stimuli ultrasonores à un milieu exempt de cellules, lequel est ensuite mélangé aux cellules. Un tel procédé de préparation d'exosomes permet l'acquisition d'exosomes possédant un effet de régénération sur les cheveux, présentant un rendement élevé sur une courte période de temps en utilisant le processus simple d'ultrasonication, à partir de cellules souches difficiles à manipuler ainsi qu'à partir de cellules somatiques qui peuvent être facilement obtenues et maintenues. Les exosomes ainsi obtenus contiennent également une grande quantité de divers facteurs. Les exosomes et une composition les contenant comprennent des quantités accrues de divers facteurs capables d'induire la régénération des cheveux. Du fait de leur structure membranaire à base de phospholipides, les exosomes peuvent facilement pénétrer dans le cuir chevelu et libérer un matériau à un taux élevé permettant d'induire efficacement la régénération des cheveux. Les exosomes sont avantageusement exempts des effets indésirables résultant de l'utilisation de médicaments synthétiques.

세포에 직간접적으로 초음파 자극을 제공하는 단계; 상기 세포 및 배지의 혼합물을 일정 시간 배양하는 단계; 및 상기 혼합물로부터 엑소좀을 분리하는 단계;를 포함하고, 상기 직접적으로 자극을 제공하는 것은 세포가 포함된 배지에 초음파 자극을 가하는 것이고, 상기 간접적으로 자극을 제공하는 것은 세포가 포함되지 않은 배지에 초음파 자극을 가한 후 상기 배지와 상기 세포를 혼합하는 것인, 모발 재생 효과를 가진 엑소좀의 제조방법을 개시한다. 이러한 엑소좀의 제조방법은, 다루기 어려운 줄기세포뿐 아니라 손쉽게 얻고 유지할 수 있는 체세포로부터도 초음파 처리라는 간단한 과정을 통해 단시간 안에 높은 수율로 모발 재생 효과를 가진 엑소좀을 수득할 수 있다. 이렇게 수득한 엑소좀은 포함된 각종 인자의 양 및 수 역시 높고, 상기 엑소좀 및 이를 포함하는 조성물은 모발 재생을 유도할 수 있는 다양한 인자가 증가되어 있으며, 엑소좀은 인지질 기반의 막구조를 가져 두피 침투가 용이하여 물질의 전달 효율이 높아 모발 재생을 효과적으로 유도할 수 있고, 합성의약품의 사용으로 나타나는 부작용이 없다는 장점이 있다.
