Abstract
(EN) A significant interferon (IFN) response is induced following treatment of CHO cells with exogenously-added type I IFN or poly I:C. Treatment of the CHO cells with poly I:C prior to infection limited the cytopathic effect from Vesicular stomatitis virus (VSV), Encephalomyocarditis virus (EMCV), and Reovirus-3 virus (Reo) in a STAT1-dependent manner. By knocking out two upstream repressors of STAT1: Gfi1 and Trim 24, the engineered CHO cells exhibited increased resistance to virus contaminations. Thus, omics-guided engineering of mammalian cell culture can be deployed to increase safety in biotherapeutic protein production.
(FR) Une réponse d'interféron (IFN) significative est induite suite au traitement de cellules CHO avec un IFN de type I ajouté de manière exogène ou avec du poly I:C. Le traitement des cellules CHO avec du poly I:C avant une infection a limité l'effet cytopathique du virus de la stomatite vésiculaire (VSV), du virus de l'encéphalomyocardite (EMCV), et du virus de réovirus-3 (Reo) d'une manière dépendante du STAT1. En supprimant deux répresseurs amont de STAT1 : Gfi1 et Trim 24, les cellules CHO manipulées par ingénierie présentaient une résistance accrue aux contaminations virales. Ainsi, l'ingénierie guidée par l'omique de la culture de cellules de mammifère peut être déployée pour augmenter la sécurité dans la production de protéines biothérapeutiques.
