Abstract

The present invention is novel Pol γ protein inhibitor compounds were identified and characterized. The compounds developed by the present invention are useful for selectively inhibiting the growth of MLH1 mutant/deficient tumor or cancer cells due to a synthetic lethal relationship between MLH1 and Pol γ. With this invention described herein were shown to directly interact with Pol γ and inhibit its biological function and cause cancer cell death and an inhibition of tumor or cancer cell growth in multiple assays.

La présente invention concerne de nouveaux composés inhibiteurs de la protéine Pol γ qui ont été identifiés et caractérisés. Les composés mis au point par la présente invention sont utiles pour inhiber sélectivement la croissance de cellules tumorales ou cancéreuses mutantes/déficientes en MLH1 en raison d'une relation létale synthétique entre MLH1 et Pol γ. Il a été démontré que la présente invention interagit directement avec Pol γ et inhibe sa fonction biologique et provoque la mort des cellules cancéreuses et une inhibition de la croissance de cellules tumorales ou cancéreuses dans de multiples essais.
