The present application claims priority for Korean patent application No. 10-2018-0072875 filed on June 25, 2018, the entire description of which is a reference for the present application. The present invention relates to a method for preparing chondrocyte pellets, comprising the steps of: (a) culturing human induced pluripotent stem cells so as to form and obtain embryoid bodies; (b) inducing the embryonic bodies obtained in step (a) to become outgrowth cells and isolating same; and (c) culturing the outgrowth cells isolated in step (b) in a pellet form. In addition, the present invention relates to: a pharmaceutical composition to be used in the treatment of arthritis, comprising the chondrocyte pellets prepared by the preparation method; or a method for preventing or treating arthritis, comprising a step of administering the chondrocyte pellets prepared by the method to an arthritis patient. Chondrocyte pellets of the present invention have a remarkably high rate of differentiation into chondrocytes, have a uniform size, and have a homogeneous differentiation degree, and a pharmaceutical composition for treating arthritis, comprising same, has an excellent cartilage regeneration effect when administered to the site of cartilage damage, and is in an injectable form, which does not require surgery, such that through a simple procedure, the pain of a patient is relieved and a continuous arthritis treatment effect can be provided even after the procedure.

La présente demande revendique la priorité pour la demande de brevet coréen n° 10-2018-0072875 déposée le 25 juin 2018 dont toute la description constitue une référence pour la présente demande. La présente invention concerne un procédé de préparation de culots de chondrocytes, comprenant les étapes suivantes : (a) culture des cellules souches pluripotentes induites humaines de manière à former et obtenir des corps embryoïdes; (b) induction des corps embryonnaires obtenus à l'étape (a) pour qu'ils deviennent des cellules d'excroissance et isolation de celles-ci; et (c) culture des cellules d'excroissance isolées à l'étape (b) sous forme de culots. De plus, la présente invention concerne : une composition pharmaceutique destinée à être utilisée dans le traitement de l'arthrite, comprenant les culots de chondrocytes préparés par le procédé de préparation; ou un procédé de prévention ou de traitement de l'arthrite, comprenant une étape d'administration des culots de chondrocytes préparées par le procédé à un patient atteint d'arthrite. Les culots de chondrocytes de la présente invention ont un taux de différenciation en chondrocytes remarquablement élevé, ont une taille uniforme, et ont un degré de différenciation homogène, et une composition pharmaceutique pour le traitement de l'arthrite, les comprenant, a un excellent effet de régénération du cartilage lorsqu'elle est administrée au site de lésion du cartilage, et se trouve sous une forme injectable, qui ne nécessite pas de chirurgie, de telle sorte que, grâce à une procédure simple, la douleur d'un patient est soulagée et un effet de traitement d'arthrite continu peut être fourni même après la procédure.

본 출원은 2018년 06월 25일 출원된 대한민국 특허출원 제10-2018-0072875호를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다. 본 발명은 (a) 인간 유도 만능 줄기세포(human induced pluripotent stem cell)를 배양하여 배아체(embryoid body)를 형성 및 수득하는 단계; (b) 상기 단계 (a)의 수득된 배아체를 돌기세포(outgrowth cell)로 유도 및 분리하는 단계; 및 (c) 상기 단계 (b)의 분리된 돌기세포를 펠렛(pellet) 형태로 배양하는 단계; 를 포함하는 연골세포 펠렛 제조 방법에 대한 것이다. 본 발명은 또한, 상기 제조 방법으로 제조된 연골세포 펠렛을 포함하는 관절염 치료에 사용하기 위한 약학적 조성물; 내지 상기 방법에 의해 제조된 연골세포 펠렛을 관절염 환자에 투여하는 단계를 포함하는 관절염 예방 또는 치료방법에 대한 것이다. 본 발명의 연골세포의 펠렛은 연골세포로의 분화율이 현저하게 높으며, 크기가 균일하고 분화 정도가 균질하며, 이를 포함하는 관절염 치료용 약학적 조성물은 이를 연골 손상 부위에 투여한 경우에 연골 재생 효과가 뛰어나고, 수술이 요구되지 않는 주사가능 형태이므로 간단한 시술로 환자의 통증을 완화할 뿐만 아니라 시술 후에도 지속적인 관절염 치료 효과를 제공할 수 있다.
