Abstract
(EN) Provided herein are methods for labeling the proteomes of cells, as well as methods for labeling proteins or populations of proteins produced by cells. In some embodiments, the methods comprise introducing variant aminoacyl-tRNA synthetases and noncanonical amino acids into cells. Also provided herein are polynucleotides encoding variant aminoacyl-tRNA synthetases that recognize noncanonical amino acids. The methods and compositions provided herein are useful for, among other things, identifying target cells and identifying biomarkers of interest.
(FR) L'invention concerne des procédés pour marquer les protéomes de cellules, ainsi que des procédés pour marquer des protéines ou des populations de protéines produites par des cellules. Dans certains modes de réalisation, les procédés comprennent l'introduction de variants d'aminoacyl-ARNt synthétases et d'acides aminés non canoniques dans des cellules. L'invention concerne également des polynucléotides codant pour des variants d'aminoacyl-ARNt synthétases qui reconnaissent les acides aminés non canoniques. Les procédés et compositions selon l'invention sont utiles, entre autres, pour identifier des cellules cibles et identifier des biomarqueurs d'intérêt.
