Abstract
(EN) Provided is a culture medium for inducing the differentiation of functional cerebral cortex cells and a method for inducing differentiation, the culture medium comprising a neural cell culture medium and a nutritional additive, the nutritional additive being one or more of SU5402, BIBF1120, IBMX and glucose. The method adds a specific factor from neural stem cells or neural precursor cells, such as an FGF or VEGF signalling pathway inhibitor or a cAMP activator, to a variety of neural cells at specific times to induce differentiation, accelerating the differentiation and maturation of the neural cells, and can obtain neural cells with primary functions and stable health by inducing differentiation from human neural precursor cells for around 7-14 days, thereby reducing manufacturing costs and shortening the production cycle.
(FR) L'invention concerne un milieu de culture destiné à induire la différenciation de cellules de cortex cérébral fonctionnelles et un procédé pour induire une différenciation, le milieu de culture comprenant un milieu de culture de cellules neurales et un additif nutritionnel, l'additif nutritionnel étant un ou plusieurs additifs parmi SU5402, BIBF1120, IBMX et le glucose. Le procédé ajoute un facteur spécifique issu de cellules souches neurales ou de cellules précurseurs neurales, telles qu'un inhibiteur de la voie de signalisation FGF ou VEGF ou un activateur d'AMPc, à diverses cellules neurales à des moments spécifiques pour induire une différenciation, accélérer la différenciation et la maturation des cellules neurales, et peut permettre d'obtenir des cellules neurales ayant des fonctions primaires et une santé stable par induction d'une différenciation à partir de cellules précurseurs neurales humaines pendant environ 7 à 14 jours, en réduisant ainsi les coûts de fabrication et en raccourcissant le cycle de production.
