(FR) SYSTÈME DE REMPLACEMENT DE L'EXPRESSION DU DOMAINE DE TYPE MUCINE DES GLYCOPROTÉINES DU VIRUS EBOLA ET DU VIRUS DE MARBURG UTILISÉ EN TANT QUE NOUVELLES APPROCHES DE VACCIN
Abstract
(EN) We have developed a series of Ebola virus envelope glycoprotein (EboGP)- based chimeric fusion proteins that are still able to maintain an efficient EboGP- mediated virus entry in various cell types including human antigen-presenting cells (APCs) while presenting large viral polypeptides, such as HIV Env v3-v5 domain (as large as 241 aa), at the apex and the sides of each EboGP monomer to elicit robust host immune responses. This invention demonstrates the feasibility of an EboGP- based chimeric fusion technology as a novel vaccine approach against different microbial pathogens, including that in human and animals, and against cancers.
(FR) La présente invention concerne une série de protéines de fusion chimériques à base de glycoprotéine d'enveloppe du virus Ebola (EboGP) qui sont encore capables de maintenir une entrée efficace du virus à médiation par EboGP dans divers types de cellules notamment des cellules présentant un antigène humain (APC) tout en présentant de grands polypeptides viraux, tels que le domaine Env v3-v5 du VIH (aussi grand que 241 aa), au niveau du sommet et des côtés de chaque monomère d'EboGP pour déclencher des réponses immunitaires solides de l'hôte. La présente invention démontre la faisabilité d'une technologie de fusion chimérique à base de EboGP en tant que nouvelle approche vaccinale contre différents pathogènes microbiens, y compris celle chez l'homme et l'animal, et contre des cancers.
