Abstract:
(EN) Stem cell maturation is a fundamental, yet poorly understood aspect of human development. A DNA methylation signature deeply reminiscent of embryonal stem cells was devised to interrogate the evolving character of multiple human tissues. The cell fraction displaying the signature was found to be highly dependent upon developmental stage (fetal vs adult) and in leukocytes, it described a dynamic transition during the first 5 years of life. Significant individual variation in the embryonic signature of leukocytes was evident at birth, in childhood, and throughout adult life. The genes denoting the signature included transcription factors and proteins intimately involved in embryonic development. The DNA methylation signature traces the developmental origin of cells and informs the study of stem cell heterogeneity in humans under homeostatic and pathologic conditions.
(FR) La maturation de cellules souches est un aspect fondamental, mais mal compris, du développement humain. Une signature de méthylation d'ADN très évocatrice des cellules souches embryonnaires a été conçue pour interroger le caractère évolutif de multiples tissus humains. Il a été trouvé que la fraction cellulaire affichant la signature est hautement dépendante du stade de développement (fœtal vs. adulte) et que, dans les leucocytes, elle décrivait une transition dynamique pendant les cinq premières années de vie. Une variation individuelle significative dans la signature embryonnaire de leucocytes était évidente à la naissance, pendant l'enfance et pendant toute la durée de vie adulte. Les gènes désignant la signature comprenaient des facteurs de transcription et des protéines intimement impliqués dans le développement embryonnaire. La signature de méthylation d'ADN trace l'origine développementale de cellules et informe l'étude de l'hétérogénéité des cellules souches chez les êtres humains dans des conditions homéostatiques et pathologiques.
