Abstract:
(EN) The present invention relates to a purity analysis method for extracellular vesicles, using size exclusion chromatography, and more specifically the present invention relates to a method of using the size-specific separation ability of size exclusion chromatography and spectroscopy, that is single or in which two or more kinds are combined, in order to quantify, from various samples, the volume of extracellular vesicles and various contaminants, simultaneously, and thereby analyze the purity, total amount and yield of extracellular vesicles. The purity analysis method for extracellular vesicles, according to the present invention, has the benefit of being capable of effective analysis whilst preserving the form or properties of the extracellular vesicles. Also, the purity analysis method for extracellular vesicles, according to the present invention, makes it possible to rapidly determine, simultaneously, the purity, total amount and yield of extracellular vesicles that have been separated using a conventional method, and can be utilized in, for example, disease diagnosis, disease treatment, multi-omics research and research into the properties of extracellular vesicles which use separated extracellular vesicles. In addition, the purity analysis method according to the present invention can also be applied to the development of a new separation method.
(FR) La présente invention concerne un procédé d'analyse de pureté pour vésicules extracellulaires, faisant appel à une chromatographie d'exclusion stérique, et plus particulièrement, la présente invention concerne un procédé d'utilisation de la capacité de séparation en fonction de la taille d'une chromatographie d'exclusion stérique et d'une spectroscopie, qui est unique ou combinant au moins deux types, afin de quantifier, dans divers échantillons, le volume de vésicules extracellulaires et divers contaminants, simultanément, et ainsi d'analyser la pureté, la quantité totale et le rendement de vésicules extracellulaires. Le procédé d'analyse de pureté pour vésicules extracellulaires, selon la présente invention, a l'avantage de pouvoir réaliser une analyse efficace tout en conservant la forme ou les propriétés des vésicules extracellulaires. De plus, le procédé d'analyse de pureté pour vésicules extracellulaires, selon la présente invention, permet de déterminer rapidement, simultanément, la pureté, la quantité totale et le rendement de vésicules extracellulaires qui ont été séparées à l'aide d'un procédé classique, et peut être utilisé, par exemple, dans le diagnostic de maladies, le traitement de maladies, la recherche multiomique et la recherche des propriétés de vésicules extracellulaires qui utilisent des vésicules extracellulaires séparées. En outre, le procédé d'analyse de pureté selon la présente invention peut également être appliqué au développement d'un nouveau procédé de séparation.
(KO) 본 발명은 크기 배제 크로마토그래피를 이용한 세포밖 소포체의 순도 분석 방법에 관한 것으로서, 보다 상세하게 본 발명은 크기 배제 크로마토그래피의 크기별 분획능과 단일 또는 2종 이상이 조합된 분광학적 분석법 (spectroscopy)을 이용하여 다양한 시료로부터 세포밖 소포체와 여러 오염 물질의 양을 동시에 정량함으로써 세포밖 소포체의 순도, 총량 및 수율을 분석하는 방법에 관한 것이다. 본 발명에 따른 세포밖 소포체 순도 분석 방법은 세포밖 소포체의 형태나 성질을 보존하면서 효율적으로 분석할 수 있다는 장점이 있다. 또한 본 발명의 세포밖 소포체 순도 분석 방법은 종래의 방법을 이용해 분리된 세포밖 소포체의 순도, 총량 및 수율을 빠른 시간 안에 동시에 결정할 수 있어, 분리된 세포밖 소포체를 이용한 질병 진단, 질병 치료, 다중 오믹스 연구 및 세포밖 소포체의 특성 연구 등에 활용이 가능하다. 또한 본 발명의 순도 분석 방법은 새로운 분리 방법을 개발하는 데도 활용이 가능하다.
