Abstract:
(EN) [Problem] To provide: a quantification method enabling selective, simple, quick, and highly sensitive amino acid quantification of target amino acids (L- and/or D-form amino acids) over a broad amino acid quantification range using an aminoacyl tRNA synthetase (AARS); and an amino acid quantification kit. [Solution] The present invention provides: a method for quantifying the amount of amino acids (L- and/or D-form amino acids) in a sample using AARS, the method being characterized in that AARS and amino acids are liberated from an once formed aminoacyl AMP-AARS complex, and are used once more to form an aminoacyl AMP-AARS complex, thereby finally producing a greater number of moles of a reaction product such as pyrophosphoric acid to be measured than the number of moles of amino acids contained in the sample; and an amino acid quantification kit for carrying out said method.
(FR) [Problème] Fournir : un procédé de quantification permettant la quantification d’acides aminés sélective, simple, rapide et très sensible d’acides aminés cibles (acides aminés de forme L et/ou D) dans une large plage de quantification d’acides aminés au moyen d’une aminoacyl-ARNt synthétase (AARS) ; et un kit de quantification d’acides aminés. [Solution] la présente invention concerne : un procédé de quantification de la quantité d’acides aminés (acides aminés de forme L et/ou D) dans un échantillon au moyen d’AARS, le procédé étant caractérisé en ce que l’AARS et les acides aminés sont libérés depuis un complexe aminoacyl-AMP-AARS formé une fois, et sont utilisés une fois de plus pour former un complexe aminoacyl-AMP-AARS, de façon à produire finalement un plus grand nombre de moles d’un produit de réaction tel que l’acide pyrophosphorique à mesurer que le nombre de moles d’acides aminés contenus dans l’échantillon ; et un kit de quantification d’acides aminés pour conduire ledit procédé.
(JA) 【課題】アミノアシルtRNA合成酵素(AARS)を用いて、測定対象のアミノ酸(L体及び/又はD体のアミノ酸)を選択的且つ簡便、短時間、高感度、広範囲なアミノ酸定量範囲を可能とする定量方法及びアミノ酸定量用キットを提供すること。 【解決手段】 AARSを用いて試料中のアミノ酸量(L体及び/又はD体のアミノ酸)を定量する方法に於いて、一度形成させたアミノアシルAMP-AARS複合体からAARS及びアミノ酸を遊離させ、それらを再度、アミノアシルAMP-AARS複合体の形成に利用することによって、最終的に、測定対象であるピロリン酸等の反応産生物を、試料中に含まれるアミノ酸より多くのモル数まで産生させることを特徴とする、前記アミノ酸定量方法、及び、該方法を実施するためのアミノ酸定量用キット。
