Abstract:
(EN) Nodulisporic acids (NAs) comprise a group of indole diterpenes known for their potent insecticidal activities; however, biosynthesis of NAs by its natural producer, Hypoxylon pulicicidum (Nodulisporium sp.) is exceptionally difficult to achieve. The identification of genes responsible for NA production could enable biosynthetic pathway optimization to provide access to NAs for commercial applications. Obtaining useful quantities of NAs using published fermentations methods is challenging, making gene knockout studies an undesirable method to confirm gene function. Alternatively, heterologous gene expression of H. pulicicidum genes in a more robust host species like Penicillium paxilli provides a way to rapidly identify the function of genes that play a role in NA biosynthesis. In this work, we identified the function of four secondary-metabolic genes necessary for the biosynthesis of nodulisporic acid F (NAF) and reconstituted these genes in the genome of P. paxilli to enable heterologous production of NAF in this fungus.
(FR) Les acides nodulisporiques (NAs) comprennent un groupe d'indole diterpènes connus pour leurs activités insecticides puissantes; cependant, la biosynthèse de NAs par son producteur naturel, Hypoxylon pulicicidum (Nodulisporium sp.) est extrêmement difficile à obtenir. L'identification de gènes responsables de la production de NA pourrait permettre une optimisation de la voie de biosynthèse pour fournir un accès à des NAs pour des applications commerciales. L'obtention de quantités utiles de NAs à l'aide de méthodes de fermentation publiées est difficile, effectuer des études d'inactivation génique est une méthode peu souhaitable pour confirmer la fonction des gènes. En variante, l'expression génique hétérologue de gènes H. pulicicidum dans une espèce hôte plus robuste comme Penicillium paxilli fournit une manière d'identifier rapidement la fonction de gènes qui jouent un rôle dans la biosynthèse de NA. Dans ce travail, nous avons identifié la fonction de quatre gènes métaboliques secondaires nécessaires à la biosynthèse de l'acide nodulisporique F (NAF) et reconstitué ces gènes dans le génome de P. paxilli pour permettre la production hétérologue de NAF dans ce champignon.
