Abstract:
(EN) Proteases limit recombinant protein production in molecular pharming since endogenous plant proteases degrade foreign protein through partial hydrolysis. Protease inhibitors are used for production of recombinant proteins in plants. Genetically modified plants expressing the protease inhibitors into their apoplast provide for high level of recombinant protein accumulation which can be isolated from the plant material, usually leaves. Three protease inhibitors are described: HsTIMP2 which is a human metalloprotease inhibitor; NbPR4 which is a homolog of CaPR4 which was recently shown to be a protease inhibitor; and NbPotI which was not known previously to be a protease inhibitor, nor was it linked to transient protein expression. These protease inhibitors are used in plants as tools for increasing desired recombinant protein expression levels. (FR) Selon la présente invention, les protéases limitent la production de protéines recombinantes en pharmaceutique moléculaire puisque les protéases végétales endogènes dégradent la protéine étrangère par hydrolyse partielle. L'invention concerne donc des inhibiteurs de protéase, utilisés pour la production de protéines recombinantes dans des plantes. Des plantes génétiquement modifiées exprimant les inhibiteurs de protéase dans leur apoplasme fournissent un niveau élevé d'accumulation de protéines recombinantes qui peuvent être isolées de la substance végétale, généralement des feuilles. Trois inhibiteurs de protéase sont décrits : HsTIMP2 qui est un inhibiteur de métalloprotéase humaine ; NbPR4 qui est un homologue de CaPR4 qui s'est récemment révélé être un inhibiteur de protéase ; et NbPotI qui n'était pas connu auparavant comme étant un inhibiteur de protéase, ni comme étant lié à une expression transitoire de protéine. Ces inhibiteurs de protéase sont utilisés dans des plantes en tant qu'outils pour augmenter les niveaux souhaités d'expression de protéines recombinantes. L'invention concerne également des constructions d'expression codant pour les inhibiteurs de protéase ainsi que les protéines recombinantes.
