Abstract:
(EN) The present invention relates to the field of biochemistry, more particularly to proteomics, more particularly to protein sequencing, even more particularly to single molecule peptide sequencing. The invention discloses means and methods for single molecule protein sequencing and/or amino acid identification using cleavage inducing agent. Said cleavage inducing agents which are not specific for one particular amino acid, cleave polypeptides step by step from the N-terminus onwards and provide information on the identity of the cleaved amino acids based on the kinetics of said reaction.
(FR) La présente invention concerne le domaine de la biochimie, plus particulièrement la protéomique, plus particulièrement le séquençage de protéines, et plus particulièrement encore le séquençage des peptides d'une seule molécule. L'invention concerne des moyens et des procédés pour le séquençage des protéines d'une seule molécule et/ou l'identification d'acides aminés au moyen d'un agent inducteur de clivage. Lesdits agents inducteurs de clivage qui ne sont pas spécifiques pour un acide aminé particulier clivent les polypeptides pas à pas à partir de l'extrémité N-terminale et fournissent des informations sur l'identité des acides aminés clivés, sur la base de la cinétique de ladite réaction.
