Abstract:
(EN) Provided are amino acid sequences of engineered decarboxylase polypeptides that are useful for catalyzing the decarboxylation of L-aspartate to produce β-alanine, and the preparation process of engineered decarboxylase polypeptides as well as reaction process under industrial-relevant conditions. Provided are also polynucleotide sequences encoding engineered decarboxylase polypeptides, engineered host cells capable of expressing engineered decarboxylase polypeptides, and methods of producing β-alanine using the engineered cells. Compared to the wild-type decarboxylase, the engineered decarboxylase polypeptide provided has better activity and stability, and overcomes the inhibition by L-aspartic acid and/or β-alanine. The use of the engineered polypeptides for the preparation of β-alanine results in higher unit activity, lower cost, and has good industrial application prospects.
(FR) L'invention concerne des séquences d'acides aminés de polypeptides modifiés de décarboxylase qui sont utiles pour catalyser la décarboxylation de L-aspartate en vue de produire de la β-alanine ainsi que le procédé de préparation de polypeptides modifiés de décarboxylase et le procédé de réaction dans des conditions industrielles pertinentes. L'invention concerne également des séquences polynucléotidiques codant pour des polypeptides modifiés de décarboxylase, des cellules hôtes modifiées capables d'exprimer des polypeptides modifiés de décarboxylase et des procédés de production de β-alanine à l'aide des cellules modifiées. Par rapport à la décarboxylase de type sauvage, le polypeptide modifié de décarboxylase selon l'invention présente une meilleure activité et une meilleure stabilité et surmonte l'inhibition par l'acide L-aspartique et/ou la β-alanine. L'utilisation des polypeptides modifiés pour la préparation de β-alanine entraîne une activité unitaire supérieure, un coût inférieur et présente de bonnes perspectives d'application industrielle.
